For anaphylaxis, serum must be obtained 15 minutes to 4 hours after onset of acute clinical symptoms. For mastocytosis, serum should be obtained during a non-acute time period.

Methodology:
Mature Tryptase: Immunoassay utilizing a monoclonal capture antibody that preferentially recognizes mature alpha and beta tryptases.

Total Tryptase: UNICAP assay uses a capture monoclonal antibody that recognizes pro, pro', and mature forms of alpha and beta tryptases.
