Minimum:
Adult/Pediatric Preferred Minimum: One 4 mL
green (sodium heparin) and one normal control in a 4 mL green (sodium
heparin) from a healthy unrelated individual.
Rejection Criteria:
Refrigerated or frozen samples or samples in transport longer than 48
hours.

Interpretive Data:
White blood cells are incubated with dihydrorhodamine 123 (DHR) and
catalase, then stimulated with Phorbol 12-Myristate 13-Acetate (PMA).
Dihydrorhodamine oxidation to rhodamine by the respiratory burst of the
cell is measured by flow cytometry.
Results are reported as the ratio of the mean channel fluorescence of
stimulated cells versus unstimulated cells, which yields a stimulation
index (SI).
Comments:
If sample shows abnormal results when stimulated, and no
control was sent, test should be resubmitted with control sample to
validate the conditions of collection, processing and
transport.
