MC540 Plus PI for Apoptosis

Reagents

  • 1mg/ml MC540 (merocyaninine 540, Sigma Chemical cat# M6390) in H2O
  • 5µl PI (propidium iodide, Sigma Chemical cat# P-4170 in PBS.

Staining

  1. Resuspend 1 x 106 cells in 0.375ml culture media.
  2. Add 125µl of PI stock (5µg/ml PI in PBS)
  3. Add 2.5µl of MC540 stock (1mg/ml MC540 in H2O
  4. Mix.
  5. Wait 1 minute.
  6. Run on flow cytometer (run MC540 in PE channel).
